Cell Wash Out

      In cell culture publications, I have seen some protocols where following a specific treatment, cells are ‘washed out‘ and ‘equilibriated’ prior to lysis and collection for biochemical examination.

      I have also seen protocols using the same treatments where the authors did not ‘wash out‘ cells, and instead just collected the cells directly after the treatment.

      When is it necessary to ‘wash out‘ cells in culture, and what exactly is the purpose of this procedure?

      Maybe there were some substances, which could interfere with subsequent processing. E.g. I have used MeOH inducible promotor in Pichia pastoris and I had to exchange media with glycerol for that with MeOH.
