How to know that cells need protein, carbohydrates, and fats to survive? We can’t see protein, so how do we know?
And how have we come to know that protein is one of the building blocks of life?

it’s because we can isolate them from the cell. There are some AAs that a cell cannot manufacture, so it needs to obtain them from the environment. If scientists do not give the cell a certain protein, it will die. If they provide it, however, it will survive. this goes with many things, including carbs and lipids.
