Making SOC solution

      Could you please let me know how to prepare SOC solution from LB, MgCl2, MgSO4, and glucose?
      I have the following recipe but I’m not sure if that’s correct:

      800 ul LB
      4 ul MgCl2 (1M)
      4 ul MgSO4 (1M)
      16 ul glucose (1M)

      My suggestions:

      • Make the LB and adjust the pH to 7. I recommend adding Tris at pH 7 to act as a buffer, maintaining the correct pH in the SOC medium as bacteria grow.
        Autoclave the LB and wait for it to cool. (Mmm, caramel!)
        Add the glucose, MgCl2, and MgSO4. (I would go for final concentrations of 10mM MgCl2, 10mM MgSO4, and 20mM glucose. This is about double the MgCl2 and MgSO4 concentrations you’ve given, but I doubt it matters.)
        Filter-sterilize (w/ 0.2um filter according to Qiagen.)
