If I anneal a DNA oligo to the end of a mRNA strand, then perform in vitro translation. My question is can this double helix stall ribosome or not? Thanks.
depends on where you put it. If you have a mature mRNA and after it you put the DNA, nothing will happen, because the stopsignal is in front of the poly-A-tail and the 3′ UTR of the mRNA.
The DNA oligo is complementary to the mRNA before the stop codon. So, what will happen. Is there any pape about this?
i think that the translation will be stopped, bacause there’s an obstacle for the ribosome. Eeventually it will fall of. But this is not based on facts, just my guess.
I don’t know if this is tried before in an experiment.
