Dear friends,
Now I am trying to clone CD36. I got the cDNA from mouse macrophage by RT-PCR (I used the kit from Invitrogen to get total cDNA and the kit from QIAGEN to amplify CD36 cDNA), then inserted it to the vector pcDNA6 from Invitrogen. After sequencing, I found there were two bases different from those of the sequence in database of NCBI. I am wondering if a gene in different mouse strain will be a little different, I mean, one or two bases are changed by mutation when they evolate. Or it is a mistake made by polymerase during the synthesis.
