For colony PCR reactions white colonies were picked from the transformation plate. I performed transformation of cloned ANO5 cDNA and the transformation mixture was plated on a LB agar plate containing ampicillin +IPTG+X-Gal. The ANO5 cDNA was cloned in the expression vector pdEYFPC1amp. No blue colonies were detected in any my transformation plates and also in the model transformation plates. Why were there no blue colonies detected? I know about the difference in picking white or blue colonies what confuses me is that NO blue colonies were present? Help?

If I have not been specific enough or maybe to broad… Please tell me and i will do my best to make it clearer. I have searched for quite a while but its all about the differences between the two and not the absence…

isn’t the blue colony the negative one?
