Hi, I;m not familiar with this technique and was wondering if someone can help out. I have a plasmid that has an ARS and my gene of interest and I need put this gene of interest in to an yeast strain to study it. Does anyone know how I can do that. More details about the technique will be very helpful. I tried to read about some yeast integration it was hard to comprehend.

what yeast do you use? I have used pPICZ and pGAPZ in Pichia, there was part of one endogenous genes, the plasmid was cut in one place and incorporated by homologous recommbination

I’m looking at Fission yeast. Its the procedeur as to how to do that is what I;m no familiar with. thanks
