      Hi,
      does anyone know what is the maximum length of DNA that can be amplified by PCR? Please post the answer if some one knows.

      may be its 1000 bp. i am not sure

      More than that! I regularly amplify over 2 kb. with good conditions and enzymes, you can go way over that. Probably up to 10 kb.

      it all depends from polymerase and PCR-method. My scientific leader amplified more than 16 kb.

      i’ve personally amplifed 8KB fragments. the problem is that regular taq has a high error rate (approx. 1 error/1000bp). Therefore, if fidelity is extremely important use hi-fi taq (hi fidelity taq)… which uses MgSO4 (instead of MgCL). Also.. the extension time has to be increased so that taq has enough time to extend….
