I have to do plasmid extraction Bacillus thuringiensis
I have heard that plasmid extraction from Gram(+) bacteria are somewhat different from that from Gram(-).
Can anyone help to tell me what these differences are?thank you,

Mostly the lysis procedure is different. Those Gram(+) are tougher than the Gram(-). Due to their cell wall. But it also depends on the species. In my lab we work with Enterococcus spp. and Listeria spp. and the latter are much harder to break than the other.

I agree with rexarunraj, addition of lysozyme is very helpful in lysis of Gram+ cells.

Yes,addition of lysozyme is very easy.Generally,I put lysozyme in solution I.This method is used in our lab to isolation plasmid from Bacillus subtilis.
