Heya,
Could you help me with this subject? I need to find out how long do I need to sonicate DNA? any suggestions any publications??
I really appreciate your help 🙂

Have you tried Google? lots of protocols available for free.

why do you want to sonicate DNA?

To shear it in small and relatively constant fragment before say cloning. You can also use a fine needle or a nebulizer. it is cheaper and less sequence specific than restriction enzyme, with break point that should be entirely random, which should give a better chance to get complete coverage for sequence, or to avoid cutting in the middle of your gene of interest.

Thank you for all replies. Unfortunately I wasn’t able to find as many protocols about sonication as I want:(
If anyone of you has knowledge or links about it I would really appreciate it. - November 24, 2010 at 7:54 pm #102450canalonParticipant
Things like:

http://www.genome.ou.edu/protocol_book/ … artII.htmlFirst and 5th link in google for DNA sonication. I fail to see what more you would need.
